烟草内源铁蛋白NtFer1 和NtFer2 的相互作用

摘 要:

以 2 个烟草铁蛋白基因全长序列(NtFer1 和 NtFer2, GenBank 登录号: AY083924 和 AY141105)为基础, 利用细菌双杂 交系统分析不同烟草铁蛋白亚基之间及相同铁蛋白亚基之间的互作关系, 并利用Northern杂交分析2个铁蛋白基因的特异 表达。结果表明, 铁蛋白基因 NtFer1 和 NtFer2 在叶片中均有表达, 同时 2 种亚基之间存在很强的互作关系, 说明在叶片中 组成铁蛋白的 24 个亚基可能有 3 种类型, 或来自单一的 NtFer1 亚基, 或来自单一的 NtFer2 亚基, 也可能来源于不同的铁蛋 白亚基。在烟草根部组织中只有铁蛋白 NtFer1 基因大量表达, 而 NtFer2 基因的表达非常微弱, 所以根部的铁蛋白大分子 可能由单一的铁蛋白 NtFer1 亚基聚合而成的。

Protein Interaction Analysis among Tobacco Ferritin NtFer1 and NtFer2

Abstract:

Based on the full length sequence of two tobacco ferritin genes (NtFer1 and NtFer2), the differential expression of ferritin genes was detected by Northern blotting, and the protein interaction among the ferritin subunits were analyzed by BacterioMatch two-hybrid assay system. Results indicated that the NtFer1 and NtFer2 gene could be higher expressed in leave, and a strong interaction between NtFer1 and NtFer2 subunits were detected, and suggested that composed of 24 subunits of ferritin may contain three assemble types: sole kind NtFer1 subunit, or sole kind NtFer2 subunit, or multi kind subunits. In the root, only NtFer1 gene was expressed, which suggested that composed of 24 subunits of ferritin in root was sole kind of NtFer1 subunit.
